L. lactis P170 process optimisation and development
If production of a given protein in the P170 proves possible in a feasibility study, yield optimisation is the natural next step in order to reduce production costs as much as possible.
Bioneer can optimise a P170-based production process to reach a commercially relevant level of protein yield. This optimisation includes genetic improvement and/or improvement of the fermentation process:
Genetic optimisation
Our P170 system comprises a collection of more than 10 expression vectors with different characteristics and a collection of more than 10 L. lactis strains selected for high level expression of recombinant proteins.
Fermentation optimisation
Optimal conditions for the fermentation of L. Lactis harbouring the production vector are primarily identified by comparing batch and fed-batch fermentation. In addition, Bioneer has in-licensed a new membrane technology (REED, reverse electro-enhanced dialysis) from Jurag Separation A/S that enables us to improve biomass yield and thereby the amount of recombinant protein by a factor of 10. This is achived by removing growth inhibitory substances from the fermentation medium.
Other commercially relevant aspects
In order to meet a client's need for an antibiotic free production system, Bioneer is about to conclude the development of an alternative non-antibiotic selection marker system that will be launced 2009.
Another commonly expressed customer need is to avoid the use of animal components in the growth medium. In response to this need, we have developed a chemically defined medium that may be further tailored to the specific requirements relevant to the recombinant protein of interest.
